About IGF-1LR3
IGF-1 LR3 (Long R3 Insulin-like Growth Factor-1) is a recombinant analog of human IGF-1 in which an N-terminal 13-amino acid extension (MFPAMPLLSLFVNG) is added and the glutamic acid at position 3 of native IGF-1 is substituted with arginine. These modifications profoundly reduce the peptide's binding affinity for all six IGF binding proteins (IGFBP-1 through IGFBP-6) — particularly IGFBP-3 — by approximately 2,000-fold, while maintaining high agonist affinity at the IGF-1 receptor (IGF-1R). The result is a significantly more potent in vitro growth factor due to the near-elimination of IGFBP-mediated buffering.
IGF-1 LR3 is widely used as a research tool in cell culture systems where native IGF-1 bioavailability is limited by endogenous IGFBPs present in serum-containing media. Research applications include IGF-1R signaling pathway characterization (IRS-1/PI3K/Akt, Ras/MAPK), comparative IGF-1R binding kinetics studies, cell proliferation assay standardization, and investigation of IGF-axis independent of IGFBP interference in defined in vitro settings.
