About HGH (Somatropin)
Human Growth Hormone (HGH; somatropin) is a 191-amino acid single-chain polypeptide hormone synthesized and secreted by somatotropic cells of the anterior pituitary gland. The mature protein contains two intramolecular disulfide bonds (Cys53-Cys165 forming a large loop and Cys182-Cys189 forming a small C-terminal loop) and adopts a four-helix bundle tertiary structure. Somatropin binds to the GH receptor (GHR) as the trigger for a sequential receptor dimerization event — first binding one GHR at site 1, then recruiting a second GHR at site 2 — that initiates JAK2/STAT5b and MAPK/PI3K intracellular signaling cascades.
Research applications of somatropin encompass GH receptor binding affinity and kinetics characterization in receptor expression systems, JAK2/STAT5b pathway activation studies in liver and muscle cell culture models, IGF-1 axis stimulation pharmacology, lipolytic and diabetogenic effects characterization in adipocyte models, and structural studies of the GH/GHR complex using biochemical and biophysical methods.
